The human microbiome is a dynamic system of microbial species that are involved in a vast array of homeostatic functions in the human body. Our findings suggest inhalation exposure to either MVE or WS result in alterations in components involved in mucosal integrity, and also microbiota profiles and diversity, which are associated with increased markers of an inflammatory response. Both WS and MVE-exposure resulted in decreased intestinal bacterial diversity, as well as alterations in microbiota profiles, including the Firmicutes: Bacteroidetes ratio at the phylum level. We observed a decrease in intestinal Muc2 and TJ proteins in both MVE and WS exposures, compared to FA controls, associated with a significant increase in MMP-9, TLR-4, and inflammatory marker expression. Microbial profiling of the intestine was done using Illumina 16S sequencing of V4 16S rRNA PCR amplicons. Immunofluorescence and RT-PCR were used to quantify the expression of IEB components and inflammatory factors, including mucin (Muc)-2, tight junction (TJ) proteins, matrix metalloproteinase (MMP)-9, tumor necrosis factor (TNF)-α, and interleukin (IL)-1β, as well as Toll-like receptor (TLR)-4. To do this, male 8 wk-old ApoE −/− mice, on a high-fat diet, were exposed to either MVE (300 μg/m 3 PM), WS (~450 μg/m 3 PM), or filtered air (FA) for 6 hr/d, 7d/wk, for 50 d. Thus, we investigated the effects of inhaled ubiquitous air-pollutants, wood-smoke (WS) and mixed diesel and gasoline vehicle exhaust (MVE) on alterations in the expression of markers of integrity, inflammation, and microbiota profiles in the intestine of atherosclerotic Apolipoprotein E knockout (ApoE −/−) mice. To date, the effects of inhaled air pollution mixtures on the intestinal epithelial barrier (IEB), and microbiota profiles are not well characterized, especially in susceptible individuals with comorbidity. The other half will be given after a 90-day probationary period.Air pollution exposure is known to contribute to the progression of cardiovascular disease (CVD) and there is increasing evidence that dysbiosis of the gut microbiome may also play a role in the pathogenesis of CVD, including atherosclerosis. He's enthusiastic to keep building upon the company's foundation and is looking forward to choosing the right person to come on board.Īccording to the company's website, if the the job requirements are met, half the reward will be provided on the technician's first day of work. In due time, he decided to start an auto service business where Integrity became the highest order of operation. “What brought me to La Pine is that I helped start a church over here, Calvary Chapel,” he said. “I was working on cars, far before I was legal to drive them, really," he said.īut working on cars wasn’t the driving factor that prompted Johnson to move to La Pine. Johnson has also had a deep interest in working on cars. The mission statement for the auto service also makes it clear that the business is not just looking for any certified technician, but one who values morals just as much as performance. "It's to be complete or whole, and by that, we mean somebody who can be of great service to the client and be very skilled at what they do," he said. He explains integrity as two-fold and shares the other part of his definition. He said Wednesday he believes that Integrity is not just about honesty and trustworthiness. Owner Jeremy Johnson is an Oregon native and family man who has made La Pine his home for the last 17 years.
